Macconkey Agar Plate

MacConkey Agar is based on the bile salt-neutral red-lactose agar of MacConkey.3 the original MacConkey medium was used to differentiate strains of Salmonella typhosa from members of the coliform group. Formula modifications improved growth of Shigella and Salmonella strains. These modifications include the addition of 0.5% sodium chloride, decreased agar content, altered bile salts, and neutral red concentrations. The formula modifications improved differential reactions between enteric pathogens and coliforms.

Salmonella Shigella agar Plate

Salmonella Shigella agar comprises of bile salts, sodium citrate, brilliant green, enzymatic digest of casein, beef extract, enzymatic digest of animal tissue, thiosulphate, ferric citrate, neutral red and agar. The inclusion of Bile Salts, Sodium Citrate and Brilliant Green serve to inhibit gram-positive, coliform organisms and inhibit swarming Proteus spp., while allowing Salmonella spp. to grow. Beef Extract, Enzymatic Digest of Casein, and Enzymatic Digest of Animal Tissue provide sources of nitrogen, carbon, and vitamins required for organism growth. Lactose serves as a carbohydrate source in Salmonella Shigella Agar. Differentiation of enteric organisms is achieved by the incorporation of lactose in the medium. Organisms which ferment lactose produce acid which, in the presence of the neutral red indicator, results in the formation of red/pink colonies. Lactose non-fermenters form colorless colonies. Sodium thiosulfate and Ferric Citrate permit detection of hydrogen sulfide by the production of colonies with black centers. Neutral red turns red in the presence of an acidic pH, thus showing fermentation has occurred.

Levine EMB Agar plate

Levine EMB Agar was developed by Levine (6,7) and is used for the differentiation of Escherichia coli and Klebsiella aerogenes and also for the rapid identification of Candida albicans. This medium is recommended for the detection, enumeration and differentiation of members of the coliform group by American Public Health Association (2,8,9). Weld (10,11) proposed the use of Levine EMB Agar, with added Chlortetracycline hydrochloride, for the rapid identification of Candida albicans in clinical specimens. A positive identification of Candida albicans can be made after 24-48 hours incubation at 35-37°C in 10% carbon dioxide atmosphere, from specimens such as faeces, oral and vaginal secretions and nail or skin scraping etc. However, the typical appearance is variable.
